The experiment used standard NA12878,Input 100 ng and enzymatic fragmentation for library preparation (QuarPrep EZ DNA Library Kit 1.0, Dynegene, NL1002), used QuarXeq Human All Exon Probes 1.0 (Dynegene, NY1001) as the panel for capture, and used QuarHyb One Reagent Kit (Dynegene, NC1003) as the capture reagent to carry out single-hybridization, 4-mixed-with-1, 6-mixed-with-1, and 8-mixed-with-1 operations, respectively.

Figure 2 showed the performances of Dynegene whole Exon 3.0 panel using the old version of QuarHyb Reagent Kit and new QuarHyb One Reagent Kit respectively. The library was prepared by the same NA12878 ultrasonic 2.0 library preparation kit and processed by two different lab technicians, A was a new hand and B was a senior one. Each sample repeated twice. According to the data above, the upgraded 2.0 reagent improved the capture efficiency, and even the data generated by new hand achieved the ideal capture rate. In addition, the data also showed that the upgraded version 2.0 reagent significantly improved the consistancy comparing to the old version of QuarHyb Reagent Kit.
