High Cloning Positive Rate with Multi-Fragment Homologous Recombination Compatibility

The QuarCam Plus Seamless Cloning Kit (Ready Mix) (Cat # NG1005) contains pre-mixed optimized recombination-enhancing factors. Multi-fragment assembly experiments were conducted using QuarCam Plus Seamless Cloning Kit (Ready Mix) (Cat # NG1005), a conventional Gibson reagent, and a competitor's reagent. After transformation and plating, colonies were picked for colony PCR. Agarose gel electrophoresis confirmed correct fragment sizes as colony PCR positive. Colony PCR-positive clones were subjected to Sanger sequencing, and clones with perfectly correct recombinant region sequences were scored as positive.

Results show that QuarCam Plus Seamless Cloning Kit (Ready Mix) (Cat # NG1005), with the added enhancing factors, significantly improves the positive clone rate, outperforming the competitor.
